Determination of Vitamin D2 Content in Ethyl Acetate Extract of Mushrooms (Volvariella volvacea L.) Using UV-VIS Spectrophotometry Method

Abstract

Mushrooms are a food source that contains vitamin D. Mushrooms are widely produced in Indonesia, especially in areas with high sunlight intensity such as Bengkulu Province, which produces palm mushrooms (Volvariella volvacea L.). These palm mushrooms grow abundantly in palm oil mill areas because they are produced from empty palm fruit bunches. Palm mushrooms are widely used by local communities as a food source. The purpose of this study was to determine the vitamin D2 content in palm mushrooms produced from palm oil plantations using UV-Vis spectrophotometry. The palm mushrooms were made into a simplisia and extracted using ethyl acetate solvent by maceration, then evaporated to obtain a thick extract. The extraction produced a yield of 1.076%. The concentration of this thick extract was then determined using UV-Vis spectrophotometry with Vitamin D2 (ergocalciferol BPFI) as a reference. From the results of testing with a UV-Vis spectrophotometer, a standard vitamin D2 solution curve was obtained with a regression coefficient (r) of 0.997 and a line equation of y = 0.0337x + 0.011. The absorbance of the sample obtained was then plotted into the line equation and its content was calculated, resulting in a content of 0.1363%. From this study, it can be concluded that palm mushrooms obtained from around palm oil plantations in Bengkulu province contain vitamin D2 at a concentration of 0.1363% or approximately 54.52 IU/gram.
