Anti-proliferative Effect of Nine Resins from Medicinal Plants of Burkina Faso

Abstract: Exudates are substances that ooze out of the pores of diseased and/or damaged tissue in plant that are well-used in flourishing international and local uses in Burkina Faso. Previously, the ethnopharmacological investigations in Center region showed that there are lot of medicinal uses like as old wounds, skin disorders diseases treatment. The current study aims to highlight the antitumoral potential of nine resins form central region. One hundred grams of each resin were placed at -80°C before extraction. The resins were grinded, and the powder was stirred twice with 1L and 500 mL of CHCl3 respectively for 24h. The antiproliferative effect was determined by using MTT colorimetric assay with chloroformic extracts from resin of each plant and the concentrations which inhibited 50% of cell proliferation (IC50) were determinated graphically. The results showed that the IC50 of A549 proliferative inhibition was form 5.78 (Lannea microcarpa) to 32.48 µg/mL (Daniellia oliveri) and U373 inhibition IC50 was from 12.90 (L. macrocarpa) to 41.12 µg/mL (Boswellia dalzielli). The Keratinocytes IC50 was form 1.57 (L. macrocarpa) to 20.4 µg/mL (Boswellia dalzielli). Interestingly, the resin extract form L. macrocarpa resin was a best source of anticancer compounds for the A549 and U373 following by Bombax costatum resin. In the contrast Commiphora africana (Burseraceae) resin was the less antiproliferation effect. The exudates resin from plants that were a source of bioactive compounds could explain of the medicinal uses like as anti-inflammatory, wound healing, and skin ailments in Burkina Faso.

Abstract: Background: Traditional Mongolian medicine Eerdun Wurile is one of the ideal drugs for cerebral stroke and is effective on treating vascular and nervous system diseases. Numerous studies have proved that Eerdun Wurile has neuroprotective effect. However, the biological mechanism of which is still unclear. Objective: To investigatethe molecular mechanism underlying the neuroprotective effect of Eerdun Wurile. Methods: The extract from Eerdun Wurile was prepared by simulated gastric fluid first and its effect on PC12 cells differentiaton and MAPK phosphorylation was tested by neurite outgrowth assay and western blotting. Also, the target of active compounds was predicted by moleculr modeling system and the content of active compounds was examined by LC-MS/MS. Result: Result showed that the extract stimulated the differentiation of PC12 cells, exhibited nerve growth factor (NGF)-like effect. Meanwhile, the extract induced the MAPK phosphorylation in PC12 cells. In-depth analysis by molecular modeling technology predicted that genipin interact with MAPK as a target. Consistent with the prediction, genipin induced the phosphorylation of MAPK in PC12 cells. Besides, genipin was detected both in the extract and Eerdun Wurile pills. Conclusion: These results proved that the extract of Eerdun Wurile has neuroprotective effect in PC12 cells, and genipin in the extract and Eerdun Wurile might be one of the active compounds that show neuroprotective effect.

Abstract: Community pharmacists are one of the most accessible of all health workers in the community, who are playing a key role in the delivering the primary health care services. Traditionally pharmacists' role is mostly confined to dispensing medication and provides public health services rarely. The current role of community pharmacists should include dispensing medicines with a prescription or without a prescription, advice patients during dispensing of prescription and non-prescription drugs, deliver drug information to health professionals, patients and to the public, and promotes health programmes. This advancement in pharmacy service demands competent pharmacists. The objective of the study was to evaluate the competency and attitude of pharmacists in the management of common ailments among community pharmacies in Adama Town, Adama, Ethiopia, 2021. A cross sectional study was conducted on 44 pharmacists working at community pharmacies in Adama Town from April 30 to July 30, 2021. Data was collected by face-to-face interview. Data was cleared, categorized, compiled, and coded before analyzed by using SPSS version 26.0. Descriptive statistics computed for the study variables and frequency distribution tables and figures was used to describe the findings. Majority of the study participants, 23 (52.3%), were males, the mean age of the participants is 33 years old. The study revealed that 26 (59.1%) were competent [mean > 0.7815, SD=0.27] while 40.9% were incompetent [mean < 0.7815]. Accordingly 50% of respondents had positive attitude (mean > 3.67) while the rest, 50% were negative attitudes. Lack of knowledge or clinical skills, lack of access to additional training programs and lack of time are the major reasons for incompetency. Majority of the community pharmacist are competent. But, 50% of the pharmacists had a negative attitude toward managements of common ailments. Reasons for incompetency such as inadequate knowledge, lack of access to training programs, inadequate time and the negative attitudes of pharmacists may be barriers for managements of common ailments. Town Health Bureaus should organize and provide in-service training and advocate managements of common ailments as one area in a continuous professional development program to pharmacists working in community pharmacies.
